Kênh kali nhạy cảm với ATP (KATP), kênh này đóng vai trò trung tâm trong việc điều hòa sự tiết insulin trong các tế bào β tuyến tụy. Đa hình Glu23Lys làm thay đổi hoạt động của kênh kali này, do đó ảnh hưởng đến nồng độ insulin máu, từ đó tác động đến các hoạt động chuyển hoá của cơ thể. Nghiên cứu cắt ngang này được thực hiện với mục tiêu xác định mối liên quan giữa đa hình Glu23Lys với hội chứng chuyển hoá ở người Việt. Đối tượng nghiên cứu gồm 2402 người từ 40-64 tuổi (436 người có hội chứng chuyển hoá và 1966 người không hội chứng chuyển hoá). Kiểu gen được xác định bằng phương pháp nhân gen đặc hiệu từng alen. Phân tích hồi quy logistic đơn biến và đa biến để xác định yếu tố nguy cơ đối với hội chứng chuyển hoá. Kết quả cho thấy tỷ lệ kiểu gen Glu/Glu, Glu/Lys và Lys/Lys ở quần thể nghiên cứu lần lượt là 44,6%, 44,0% và 11,4% và theo luật cân bằng Hardy-Weinberg (p=0,55). Người có kiểu gen đồng hợp tử Lys/Lys có nguy cơ bị hội chứng chuyển hoá cao hơn 1,36 lần (95%CI=1,01-1,85) so với người có kiểu gen Glu/Glu và Glu/Lys. Người mang kiểu gen dị hợp tử Glu/Lys giảm cơ nguy cơ bị hội chứng chuyển hoá hơn nhóm còn lại (OR= 0,79; 95%CI=0,63-0,99). Nghiên cứu cho thấy đa hình Glu23Gly trên gen KCNJ11 có liên quan đến nguy cơ mắc hội chứng chuyển hoá ở người Việt.

The ATP-sensitive K channel plays a central role in regulation of insulin secretion in β cells of pancreas. As the Glu23Lys polymorphism in KCNJ11 gene changes the channel activity, it influences blood insulin level. Thereby, it contributes into metabolic activities in human body. A cross-sectional study aimed to identify the relationship between the Glu23Lys polymorphism and metabolic syndrome (MetS) in Vietnamese population. The study subjects included 2402 habitants aged 40-64 years (432 subjects with MetS and 1966 subjects without MetS). Genotypes were identified using Allele Specific Primer-PCR method. Univariate and multivariate logistic regressions were performed to investigate associations between the polymorphism and MetS. The results showed that the percentages of Glu/Glu, Glu/Lys and Lys/Lys in the Vietnamese population were respectively 44.6, 44.0 and 11.4%; and they were in Hardy-Weinberg Equilibrium (p=0.55). Carriers with Lys/Lys had 1.36-fold risk (95%CI=1.01-1.85) to MetS as comparison to those with Glu/Glu and Glu/Lys. Carriers with Glu/Lys had lower risk of MetS (OR= 0,79; 95%CI=0,63-0,99) compared to the others. In summary, the study suggests that the KCNJ11 Glu23Lys polymorphism is significantly associated with MetS in the Vietnamese population.
